Danai Simantiri wurde 1997 in Athen geboren. Nach ihrem deutschen und griechischen Abschluss an der Deutschen Schule Athen sammelte sie Bühnenerfahrung in der ersten professionellen Musical Hochschule Griechenlands, P.M.T.P. by Sia Koskina. 2017 zog sie nach Berlin und nahm intensiv Unterricht in Gesang, Tanz und Schauspiel. Im August 2018 fing sie eine studienvorbereitende Ausbildung (SVA) für Musical an der Paul-Hindemith-Musikschule Berlin Neukölln an. Seit März 2019 studiert sie Musical an der Theaterakademie August Everding. Zu sehen war sie im Sommer 2021 am Staatstheater Augsburg als Jane im Musical „Chicago“.
